If your trying to isolate or extract vocals using Audacity this is exactly how you do it. You will want to make sure you have both the quality version of the song and instrumental so that you can extract and make the best acapella from your mp3. This process is called a waveform match and track inversion. You will import both your song and instrumental into audacity and using (command+1 for mac users) you can zoom in to see your waveforms up close. You want to make sure that the peaks of your waveforms match up perfectly before you invert the instrumental track. What you are essentially doing is using the song and inverted instrumental to cancel out the beat so to isolate and extract just the vocals of your song.  If your song and instrumental do not match perfectly you will get background feedback in your acapella.  This is a video tutoial by the how to mac.
